The protein p21WAF1/Cip1 inhibits the activity of several cyclin/cyclin-dependent kinase complexes and blocks cell-cycle progression (1). In tumor cells that have lost the p53 protein, or contain an altered form of p53, p21WAF1/Cip1 levels are dramatically reduced or totally absent (2). Because p21WAF1/Cip1 appears to mediate several of the growth-regulatory functions of p53, its expression would be predicted to reflect the functional status of p53 more precisely than p53 accumulation. The expression of p21WAF1/Cip1 has been studied by immunohistochemical methods in a wide range of human tumors. For optimal results on formalin-fixed, paraffin-embedded specimens, heat-induced epitope retrieval in 10 mmol/L Tris buffer, 1 mmol/L EDTA, pH 9.0, high pH, is recommended.

Monoclonal Mouse Anti-Human p21WAF1/Cip1, Clone SX118, is intended for use in immunocytochemistry. The antibody labels cells expressing p21WAF1/Cip1 may be an aid, e.g. in the investigation of gastric carcinoma, where loss of p21WAF1/Cip1 expression may contribute to increased proliferative activity (1). Differential identification is aided by the results from a panel of antibodies. Interpretation must be made within the context of the patient's clinical history and other diagnostic tests by a qualified pathologist.
In immunoprecipitates of cell extract from the human osteosarcoma cell line SAOS-2 transfected with a plasmid expressing p21WAF1/Cip1, the antibody labels a single band corresponding to p21WAF1/Cip1. The corresponding immunoprecipitates from non-transfected SAOS-2 cells were not labelled by the antibody (7).
The epitope recognized by the antibody is localized to the last 20 amino acids of p21WAF1/Cip1 (7).
As demonstrated by Western blotting, the antibody labels the p21WAF1/Cip1 - equivalent protein in mouse (7).
