Reacts with nucleophosmin (NPM) and its mutated counterpart NPMc. NPM is predominantly localized in the nucleus of cells in most tissues.
However, NPMc is aberrantly accumulated in the cytoplasm of leukemic blasts in a large subgroup of acute myeloid leukemia (AML) cases with a normal karyotype (1-2). In contrast nuclear and cytoplasmic labeling of leukemic blast cells is only observed in ~10% of AML cases with an abnormal karyotype, and none of these cases represent specific genetic abnormalities commonly observed in primary AML (1). Immunohistochemical detection of the NPMc phenotype is a tool for diagnosing AML carrying NPM1 exon 12 mutations and it may represent a simple and valuable indicator for lack of recurrent chromosomal abnormalities in primary AML (3).

FLEX Monoclonal Mouse Anti-Human Nucleophosmin, Clone 376, Ready-to-Use (Link), is intended for use in immunohistochemistry together with Autostainer Link instruments. This antibody is useful for the identification of both wild-type nucleophosmin (NPM) and the mutated counterpart NPMc+. Nucleophosmin is localized in the nuclei of normal cells in multiple tissues, but is aberrantly accumulated in the cytoplasm of leukemic blasts in a large subgroup of acute myeloid leukemia cases with a normal karyotype (1-3). The clinical interpretation of any staining or its absence should be complemented by morphological studies using proper controls and should be evaluated within the context of the patient's clinical history and other diagnostic tests by a qualified pathologist.
