BCL6 Protein
Clone PG-B6p
The BCL6 gene encodes a 706 amino acid nuclear protein of the Kruppel-type zinc finger protein.
It is rearranged in about 30% of diffuse large B-cell lymphomas, and is expressed predominantly in normal germinal centre B cells and related lymphomas (1). The antibody gives a strong nuclear labeling of BCL6 protein in follicular lymphomas, diffuse large B-cell lymphomas, Burkitt's lymphomas, and nodular, lymphocyte-predominance Hodgkin's disease. The PG-B6p antibody is useful for differentiating proliferation centres in B-CLL (BCL2+/BCL6-) from trapped germinal centres (BCL2-/BCL6+) in mantle cell lymphomas, and for identifying neoplastic cells in cases of nodular, lymphocyte-predominance Hodgkin's disease. BCL6 protein is not expressed in B-CLL, hairy cell leukemia, mantle cell and marginal-zone derived lymphomas (2, 3).

Monoclonal Mouse Anti-Human BCL6 Protein, Clone PG-B6p, is intended for use in immunohistochemistry. The antibody labels cells expressing BCL6 protein and is a useful tool for differentiating proliferating centres in B-cell chronic lymphocytic lymphomas (BCL2+/BCL6-) from trapped germinal centres in mantle cell lymphomas (BCL2-/BCL6+), and for identifying neoplastic cells in cases of nodular, lymphocyte-predominance Hodgkin's disease (1). The clinical interpretation of any staining or its absence should be complemented by morphological studies using proper controls and should be evaluated within the context of the patient's clinical history and other diagnostic tests by a qualified pathologist.
In Western blotting of total lysates of Bjab cells and BCL6 transfected EB3 cells, or control transfected EB3 cells, the antibody did not label any bands, suggesting lack of reactivity with denatured BCL6 protein (1).
As demonstrated by immunohistochemistry, the antibody cross-reacts with the BCL6-equivalent protein in various mammals, including cow, rabbit, rat, sheep, and swine. No cross-reactivity was observed in chicken and pigeon (1).

The protein concentration between lots may vary without influencing the optimal dilution. The titer of each individual lot is compared and adjusted to a reference lot to ensure a consistent immunohistochemical staining performance from lot-to-lot.

FLEX Monoclonal Mouse Anti-Human BCL6 Protein, Clone PG-B6p, Ready-to-Use (Link), is intended for use in immunohistochemistry together with Autostainer Link instruments. This antibody is useful for classification of diffuse large B-cell lymphomas, follicular lymphomas and Burkitt's lymphomas (1).
