Cytokeratin 18
Clone DC 10
Reacts with the 45 kDa protein corresponding to cytokeratin 18. The antibody labels a large number of simple epithelia, including many ductal and glandular epithelia.
The antibody labels tumor cells of epithelial origin, whereas tumor cells of non-epithelial origin such as glioma, melanoma and osteosarcoma, are not labeled. In vascular tumors, the antibody labels endothelial cells in epithelioid hemangioendotheliomas.

Liver stained with FLEX Anti-Cytokeratin 18, Code IR618/IS618.
Liver. The bile duct epithelial cells show a moderate to strong cytoplasmic staining reaction, whereas the liver cells show a weak to moderate staining reaction.The staining reaction can be heterogeneous with the strongest staining in the periportal zones. Kupffer cells should be negative.
Recommended control for IR618/IS618. Liver.
Merkel cell carcinoma. The majority of the neoplastic cells show a distinct cytoplasmic and focally dot-like staining reaction.
Clinical application for IR618/IS618. For identification of some epithelial tumors like Merkel cell carcinoma, seminoma and epithelioid hemangioendothelioma.
Seminoma. Several neoplastic cells show a focal and distinct cytoplasmic staining reaction - in some areas the staining may be more dot-like. The majority of the neoplastic cells are negative.
Clinical application for IR618/IS618. For identification of some epithelial tumors like Merkel cell carcinoma, seminoma and epithelioid hemangioendothelioma.

Monoclonal Mouse Anti-Human Cytokeratin 18, Clone DC 10, is intended for use in immunohistochemistry. The antibody labels epithelial cells in those epithelia expressing cytokeratin 18, and hence the antibody is a useful tool for the differentiation and identification of epithelial tumors (1) and epithelioid hemangioendotheliomas (2). Differential identification is aided by the results from a panel of antibodies. Interpretation must be made within the context of the patient´s clinical history and other diagnostic tests by a qualified pathologist.
In immunocytochemistry on human cultured cell lines, the antibody labels tumor cells of epithelial origin, whereas normal fibroblasts and tumor cells of non-epithelial origin such as glioma, melanoma and osteosarcoma, are not labeled (6).
As demonstrated by immunocytochemistry, the antibody cross-reacts with the CK 18-equivalent protein in monkey (7). No cross-reactivity with the CK 18-equivalent protein is observed in cow, dog, hamster, mouse, rat, sheep and swine (6).

The protein concentration between lots may vary without influencing the optimal dilution. The titer of each individual lot is compared and adjusted to a reference lot to ensure a consistent immunohistochemical staining performance from lot-to-lot.

FLEX Monoclonal Mouse Anti-Human Cytokeratin 18, Clone DC 10, Ready-to-Use (Link), is intended for use in immunohistochemistry together with Autostainer Link instruments. This antibody labels epithelial cells in those epithelia expressing cytokeratin 18, and hence the antibody is a useful tool for the differentiation and identification of epithelial tumors (1) and epithelioid hemangioendotheliomas (2). Antibodies to cytokeratin 18 may also be useful for the identification of seminomas (3). The clinical interpretation of any staining or its absence should be complemented by morphological studies using proper controls and should be evaluated within the context of the patient's clinical history and other diagnostic tests by a qualified pathologist.
