Monoclonal Mouse Anti-Human Chromogranin A, Clone DAK-A3, is intended for use in immunocytochemistry. The antibody labels chromogranin A present in secretory granules of endocrine cells. Chromogranin A is a marker for neuroendocrine differentiation and antibodies to chromogranin A are a useful tool for the identification of neuroendocrine-derived tumours. Differential identification is aided by the results from a panel of antibodies. Interpretation must be made within the context of the patient's clinical history and other diagnostic tests by a qualified pathologist.
In immunocytochemistry the antibody shows a staining pattern essentially as that of an established polyclonal antibody to chromogranin A, Dako Code A0430, when tested on 84 different tumours and 30 different normal tissues. The labelling corresponds to the known distribution of chromogranin A. In addition, labelling of melanin and lipofucsin pigment was observed with both antibodies as also with an unrelated monoclonal antibody to chromogranin A, Clone LK2H10, after demasking of antigens with citrate buffer.
