Monoclonal Mouse Anti-Epidermal Growth Factor Receptor (anti-EGFR) is intended for laboratory use to identify qualitatively by light microscopy epidermal growth factor receptor positive cells in normal and neoplastic tissues using immunohistochemical (IHC) test methods. The clinical interpretation of any positive staining or its absence should be complemented by morphological and histological studies with proper controls. Evaluations should be made within the context of the patient's clinical history and other diagnostic tests by a qualified individual.
A combination immunization protocol was employed using microsomal membrane preparations of HC2 20 d2 xenograft cells and a 14 amino acid EGFRvIII-specific synthetic peptide, Pep 3 conjugated to KLH (1).
In Western blots of A431 cell extracts, monoclonal anti-human EGFR, clone H11 (anti-EGFR) recognizes the Mr 170 kD wild-type EGFR and the Mr 145 kD deletion mutant form of the receptor (EGFRvIII). In an analytical flow cytometric assay, anti-EGFR was demonstrated to strongly label the surface of a homogeneous population of A431 cells which overexpress the normal EGFR by a factor of 20-50 fold. Anti-EGFR also positively stained cells from the EGFRvIII-transfected HC2 20 d2 cell line (1).
