Acute myeloid leukemia (FFPE) stained with FLEX Anti-CD68, Code IR609/IS609.
Acute myeloid leukemia (FAB type M5). The neoplastic cells show a distinct granular cytoplasmic staining reaction.
Clinical application for IR609/IS609. For identification of macrophages, other members of the mononuclear phagocyte lineage, and neoplasm of myeloid and macrophage/monocyte origin.

Cerebellum. The microglial cells show a weak to moderate cytoplasmic staining reaction, while the other cell types are negative.

Monoclonal Mouse Anti-Human CD68, Clone KP1, is intended for use in immunohistochemistry. The antibody labels macrophages and other members of the mononuclear phagocyte lineage and is a useful tool for the identification of neoplasms of myeloid and macrophage/monocyte origin (1). Differential identification is aided by the results from a panel of antibodies. Interpretation must be made within the context of the patient's clinical history and other diagnostic tests by a qualified pathologist.
SDS-PAGE analysis of immunoprecipitates formed between the antibody and 125I-labelled lysates from human spleen with B-cell lymphoma rich in macrophages shows reaction with a 110 kDa polypeptide, corresponding to CD68 (4).
In Western blotting of extracts of lung, spleen and U937 cells, diffuse 110, 70 and 40 kDa bands were detected when using reducing conditions. Under non-reducing conditions the spleen extract showed an additional 220 kDa band (4).

The protein concentration between lots may vary without influencing the optimal dilution. The titer of each individual lot is compared and adjusted to a reference lot to ensure a consistent immunohistochemical staining performance from lot-to-lot.
