Cytokeratin 5/6
Clone D5/16 B4
Reacts strongly with cytokeratins 5 and 6 and weakly with cytokeratin 4. The antibody does not cross-react with cytokeratins 1, 7, 8, 10, 13, 14, 18 and 19.
It labels mesothelioma, and epithelial basal cells in prostate and tonsil. No reactivity with other mesodermally derived tissues, such as muscle and connective tissues, has been observed. For optimal results on formalin-fixed, paraffin-embedded specimens, heat-induced epitope retrieval using high pH buffer is recommended.
Cytokeratin 5/6 is expressed at low levels in adenocarcinomas, whereas it is present at high levels in epithelioid mesotheliomas, and the antibody is, therefore, very useful in the differentiation between these types of cancer.

Squamous cell carcinoma of the lung (FFPE) stained with FLEX Anti-Cytokeratin 5/6, Code IR780/IS780.
Squamous cell carcinoma of lung. The majority of the neoplastic cells show a strong cytoplasmic staining reaction.
Clinical application for IR780/IS780. For differential diagnosis of poorly differentiated squamous cell carcinoma vs. adenocarcinoma, epithelioid mesothelioma vs. lung carcinoma, ductal hyperplasia vs. ductal carcinoma in situ of the breast, and prostate carcinoma vs. its benign mimics.
Prostate hyperplasia and prostate carcinoma. The normal and benign glands show a distinct cytoplasmic staining reaction in the basal cells.
Clinical application for IR780/IS780. For differential diagnosis of poorly differentiated squamous cell carcinoma vs. adenocarcinoma, epithelioid mesothelioma vs. lung carcinoma, ductal hyperplasia vs. ductal carcinoma in situ of the breast, and prostate carcinoma vs. its benign mimics.
Tonsil. The squamous epithelial cells show a moderate to strong cytoplasmic staining reaction. The staining reaction is seen in all cell layers.
Recommended control for IR780/IS780. Tonsil/Prostate.
Prostate. The basal cells show a moderate cytoplasmic staining reaction with no or only focal staining of the secretory cells.

Monoclonal Mouse Anti-Human Cytokeratin 5/6, Clone D5/16 B4, is intended for use in immunohistochemistry. Antibodies to cytokeratin 5/6 (CK 5/6) have been found valuable for the distinction between low differentiated squamous cell carcinoma and adenocarcinoma. Furthermore, the expression of CK5 by mesothelium makes this antibody valuable in the differentiation between epithelioid mesothelioma and lung carcinoma (1) when used together with other antibodies against mesothelioma markers, such as calretinin and thrombomodulin (2). Anti-CK 5/6 has also been found useful in the differential diagnosis of atypical proliferations of the breast (3). Differential identification is aided by the results from a panel of antibodies. Interpretation must be made within the context of the patient's clinical history and other diagnostic tests by a qualified pathologist.

The protein concentration between lots may vary without influencing the optimal dilution. The titer of each individual lot is compared and adjusted to a reference lot to ensure a consistent immunohistochemical staining performance from lot-to-lot.
