Myeloperoxidase
The antigen has been isolated from primary granulocytes (1). The antibody reacts with neutrophil granulocytes and monocytes in blood and with precursors of granulocytes in the bone marrow.
The antibody is valuable for discrimination between lymphoid and myeloid leukemias and as an aid in the classification of neoplastic tissue, i.e. myeloblasts and immature myeloid cells of acute myelogenous leukemia, progranulocytic leukemia, monomyelocytic leukemia, erythroleukemia and myeloblastoma (2) .

Acute myeloid leukemia (FFPE) stained with FLEX Anti-Myeloperoxidase, Code IR511/IS511.
Acute myeloid leukemia (M4). A strong granular cytoplasmic staining reaction is seen in part of the neoplastic cells, and a weak or absent staining is observed in the monocytes.
Clinical application for IR511/IS511 For differential diagnosis between lymphoid and myeloid tumors as myeloid tumors are generally positive and lymphoid tumors negative.
Liver. The neutrophils and Kupffer cells show a moderate to strong staining reaction. Hepatocytes should be negative or show a weak staining reaction.
Recommended control for IR511/IS511. Liver/Tonsil.
Tonsil. The neutrophils and eosinophils show a moderate to strong granular cytoplasmic staining reaction.

Polyclonal Rabbit Anti-Human Myeloperoxidase is intended for use in immunohistochemistry. The antibody labels myeloperoxidase-positive cells in normal and neoplastic tissues and is a useful tool for the discrimination between lymphoid and myeloid disorders. However it is important that the antibody is not used alone but together with a panel of other antibodies (1, 2). Interpretation must be made within the context of the patient's clinical history and other diagnostic tests by a certified professional.
In crossed immunoelectrophoresis using 12.5 µL antibody per cm2 gel area against 2 µL myeloperoxidase only one precipitate corresponding to myeloperoxidase appears. Staining: Specific staining for peroxidase. With 2 µL of human plasma, no precipitate appears. Staining: Coomassie Brilliant Blue.
In formalin-fixed, paraffin-embedded human tonsil, and when using a sensitive streptavidin-biotin-based visualization system, the antibody as expected labels only rare neutrophil granulocytes.
As demonstrated by immunocytochemistry, the antibody cross-reacts with the myeloperoxidase-equivalent protein in cow, horse, rat and swine.

FLEX Polyclonal Rabbit Anti-Human Myeloperoxidase, Ready-to-Use (Link), is intended for use in immunohistochemistry together with Autostainer Link instruments. The antibody labels myeloperoxidase-positive cells in normal and neoplastic tissues and is a useful tool for the discrimination between lymphoid and myeloid disorders.
