Labels myocardial, skeletal and smooth muscle cells as well as myoepithelial cells.
The antibody recognizes rhabdomyosarcomas, leiomyomas and leiomyosarcomas, and also reacts with 'myofibroblasts' in the stroma of certain tumors including many carcinomas.

Leiomyosarcoma. The majority of the neoplastic cells show a moderate to strong staining reaction.
Clinical application for IR700/IS700. For identification of soft tissue tumors with muscle differentiation, i.e. leiomyoma, leiomyosarcoma and rhabdomyosarcoma.
Rhabdomyosarcoma. The majority of the neoplastic cells show a weak to moderate staining reaction.

Colon. All the smooth muscle cells in vessel walls, lamina muscularis mucosa and myoepithelial cells around colonic crypts show a moderate to strong staining reaction. No staining reaction should be seen in the epithelial cells.

Tongue. The myoepithelial cells of the mucous/salivary glands show a weak to moderate staining reaction.

Intended Use

Monoclonal Mouse Anti-Human Muscle Actin, Clone HHF35, is intended for laboratory use to identify qualitatively by light microscopy an epitope present on muscle actin in normal and neoplastic tissues using immunohistochemical (IHC) test methods. Clone HHF35 has been demonstrated to be a reliable marker for soft tissue tumors with muscle differentiation, i.e. leiomyomas (LM), leiomyosarcomas (LMS), and rhabdomyosarcomas (RMS) (2). The clinical interpretation of any positive staining or its absence should be complemented by morphological and histological studies with proper controls. Evaluations should be made within the context of the patient's clinical history and other diagnostic tests by a qualified individual.

Specificity
Actin does not react with the α-actin of non-muscle (endothelial cells) sources (1). Gel electrophoresis and immunoblots show the specificity of HHF35 to be for the α- and (-actin isotypes of skeletal, cardiac and smooth muscle (1).
Reagent Provided
Monoclonal mouse antibody provided in liquid form as tissue culture supernatant in 0.05 mol/L Tris-HCl, pH 7.2 and 0.015 mol/L sodium azide. This product contains stabilizing protein.

Specificity
In Western blotting of purified rabbit skeletal muscle actin, extracts of monkey aorta, uterus, diaphragm and heart, the antibody labels 42 kDa protein corresponding to muscle actin of α and γ-actin isotypes, but fails to react with the α-actin of non-muscle (endothelial cells) sources (4)
