Reacts with the BCL2 oncoprotein encoded by a gene involved in the t(14;18) chromosomal translocation. The BCL2 oncoprotein plays a central role in apoptosis.
This antibody has found numerous applications in studies of apoptosis, e.g. in hematological malignancies and other malignant diseases (1, 2). The antibody may be useful in the classification of follicular lymphomas and various diffuse lymphoproliferative diseases, and for differential identification of diffuse large B-cell lymphoma versus Burkitt lymphoma/leukemia (3).

Follicular lymphoma. The majority of the neoplastic cells show a distinct cytoplasmic staining reaction.
Clinical application for IR614/IS614.. Primarily for identification of follicular lymphoma. Various B- and T-cell lymphoproliferative diseases and some diffuse large B-cell lymphomas are BCL2 positive. Burkitt lymphoma/leukemia is generally negative for BCL2.
Burkitt lymphoma/leukemia. The neoplastic cells are negative for BCL2 and only benign T cells are stained.
Clinical application for IR614/IS614. Primarily for identification of follicular lymphoma. Various B- and T-cell lymphoproliferative diseases and some diffuse large B-cell lymphomas are BCL2 positive. Burkitt lymphoma/leukemia is generally negative for BCL2.
Tonsil. The scattered T cells in the germinal centers and the mantle zone B cells show a moderate to strong cytoplasmic staining reaction. The germinal center B cells are negative.

Monoclonal Mouse Anti-Human BCL2 Oncoprotein, Clone 124, is intended for use in immunohistochemistry. The antibody labels cells expressing BCL2 oncoprotein. Positive results aid in the classification of follicular lymphomas and various diffuse lymphoproliferative diseases. The clinical interpretation of any staining or its absence should be complemented by morphological studies using proper controls and should be evaluated within the context of the patient's clinical history and other diagnostic tests by a qualified pathologist.
The antibody labels the myeloid leukaemic cell lines, HL-60 (promyelocytic), KG1 (myeloblastic), GM-1 (monoblastic) and K562 (erythromyeloid) (5).

The protein concentration between lots may vary without influencing the optimal dilution. The titer of each individual lot is adjusted to ascertain comparable lot-to-lot immunohistochemical staining performance with a reference lot.
