Kappa Light Chains
These reagents have been produced in a manner that ensures a particularly wide specificity for kappa-chains.
The specificity is directed against surface as well as hidden determinants and has been ascertained by gel precipitation techniques and immunohistochemistry. This antibody is useful for the identification of plasma cells and related lymphoid cells containing kappa light chains, and it is a useful tool for the classification of patients with monoclonal gammopathies and amyloidosis. Additionally, the antibody may be used for distinguishing neoplastic monoclonal proliferation from reactive hyperplasia of B cells.

B-cell lymphoma (FFPE) stained with FLEX Anti-Kappa Light Chains, Code IR506/IS506.
B-cell lymphoma. Most neoplastic cells show a distinct membranous and cytoplasmic staining reaction.
Clinical application for IR506/IS506 For identification of plasma cells and lymphoid cells containing kappa light chains, and for classification of kappa light chain restriction for monoclonal gammopathies and amyloidosis.
Plasmacytoma. The tumor cells show a strong cytoplasmic staining reaction.

Tonsil. B cells in the mantle zone show a weak to moderate membranous staining reaction, while the B lymphocytes in the germinal center are weakly demonstrated. Plasma cells and immunoblasts in the germinal center show a moderate to strong cytoplasmic staining.

F0198 is intended for demonstration of kappa light chains in tissues, and may also be used for other immunofluorescencetechniques (1-3). Interpretation of results must be made within the context of the patient's clinical history and other diagnostic tests by a certified professional.
In crossed immunoelectrophoresis, only the kappa precipitation arch appears when using 37.5 µL antibody per cm2 gelarea against 2 µL human plasma. Staining: Coomassie Brilliant Blue.

Antibody titre: 200 mg/L (mg of antigen to be added to 1 L of antibody to reach equivalence point).
F/P ratio: E495 nm/E278 nm = 0.65 ± 0.05 corresponding to a molar FITC/protein ratio of 2.5.

FLEX Polyclonal Rabbit Anti-Human Kappa Light Chains, Ready-to-Use (Link), is intended for use in immunohistochemistry together with Autostainer Link instruments. This antibody is useful for the identification of plasma cells and related lymphoid cells containing kappa light chains, and it is a useful tool for the classification of patients with monoclonal gammopathies (2) and amyloidosis (3, 4). Additionally, the antibody may be used for distinguishing neoplastic monoclonal proliferation from reactive hyperplasia of B cells (5-7). The clinical interpretation of any staining or its absence should be complemented by morphological studies using proper controls and should be evaluated within the context of the patient's clinical history and other diagnostic tests by a qualified pathologist.
Specificity tests; staining with Coomassie Brilliant Blue:
Double immunodiffusion: No reaction with lambda or gamma chains is seen when using 15 µL of concentrated antibody against 15 µL of lambda IgG.
Crossed immunoelectrophoresis: Only kappa-related precipitates appear when using 12.5 µL concentrated antibody per square cm gel area against 2 µL human plasma or 2 µL of a pool of concentrated urines from patients with tubular proteinuria.
