Calretinin is a 32 kDa member of the superfamily of calcium-binding proteins.
It is abundantly expressed in central and peripheral neural tissues, particularly in the retina and in the neurons of the sensory pathways, and calretinin may play an important role in the survival of nerve cells during disturbances in calcium homeostasis. Calretinin is also expressed by both normal and neoplastic mesothelial cells, and it has been suggested as a useful marker for the identification of malignant mesotheliomas of the epithelial type and for the differentiation of these malignancies of lung adenocarcinoma.

Epithelial mesothelioma. The majority of the neoplastic cells show a distinct nuclear and cytoplasmic staining reaction.
Clinical application for IR627/IS627. Primarily for identification of mesothelial cells and malignant mesothelioma of the epithelial type and in the differential diagnosis of mesothelioma vs. primary/metastatic lung or ovarian adenocarcinoma.
Granulosa cell tumor. The majority of the neoplastic cells show a distinct nuclear and cytoplasmic staining reaction.
Clinical application for IR627/IS627. Primarily for identification of mesothelial cells and malignant mesothelioma of the epithelial type and in the differential diagnosis of mesothelioma vs. primary/metastatic lung or ovarian adenocarcinoma.
Colon. The ganglion cells show a strong staining reaction and the axons show a weak to moderate staining reaction. Peripheral macrophages should also show a nuclear and cytoplasmic staining reaction. No staining should be seen in the epithelial cells.
Recommended control for IR627/IS627. Appendix/Colon.

Calretinin is expressed by both normal and neoplastic mesothelial cells and is a useful marker for the identification of malignant mesotheliomas of the epithelial type and for the differentiation of these malignancies from metastases of lung adenocarcinoma (1-3). Differential identification is aided by the results from a panel of antibodies. Interpretation must be made within the context of the patient's clinical history and other diagnostic tests by a qualified pathologist.

From the human malignant mesothelioma cell line, Mero-41 (8, 9), the gene encoding calretinin was amplified by PCR and expressed as recombinant protein in E. coli. The purified protein was used for immunization.
Specificity
In immunocytochemistry, the antibody specifically labels calretinin.
Reagent Provided
Monoclonal mouse antibody provided in liquid form as cell culture supernatant dialysed against 0.05 mol/L Tris/HCl, pH 7.2, and containing 15 mmol/L NaN3.

The protein concentration between lots may vary without influencing the optimal dilution. The titer of each individual lot is compared and adjusted to a reference lot to ensure a consistent immunohistochemical staining performance from lot-to-lot.
