Thymidylate Synthase

Thymidylate synthase (TS) is a key enzyme in the synthesis of DNA. TS, along with methyl donor 5,10-methylenetetrahydrofolate, catalyzes the methylation of deoxyuridine monophosphate (dUMP) to deoxythymidine monophosphate (dTMP) to generate the thymine nucleotides necessary for DNA biosynthesis. TS is also a target for fluorinated pyrimidine drugs such as 5-fluorouracil (5-FU). The active metabolite of 5-FU, FdUMP, competes with dUMP for the active binding site on the TS enzyme. FdUMP forms a ternary complex with TS and 5,10-methylenetetrahydrofolate, thereby inhibiting the enzyme. High levels of TS expression has been associated with resistance to 5-FU-based chemotherapy in a variety of cancers, including rectal, breast, colorectal, gastric, head and neck, esophageal, and pancreatic cancer.

Intended Use
For in vitro diagnostic use.

This antibody is intended for laboratory use to identify qualitatively by light microscopy thymidylate synthase expressing cells in normal and neoplastic tissues using immunohistochemical (IHC) test methods. Differential identification is aided by the results from a panel of antibodies. Interpretation must be made within the context of the patient's clinical history and other diagnostic tests by a qualified pathologist.

Purified recombinant human thymidylate synthase (1).
Specificity
Monoclonal mouse anti-TS recognizes recombinant TS in ELISA, immunoprecipitation and Western blot assays (1). Anti-TS has been shown to identify a 36 kD protein in Western blots of colon cancer cell lines HCT 116, NCI H630, SNU-C4 and SNU-C1 and breast cancer cell line MCF-7 (1,11). Western blot analysis of 5FU-resistant cancer sublines NCI H630R1, NCI H630R10, MCF-Ad5 and MCF-Ad10 demonstrated a significant increase in TS compared to the parent cell lines (11). Epitope mapping studies using overlapping peptides and peptides with sequential truncation have demonstrated that TS106 binds in the 10-amino acid region of P133-F142, and that residues F137, W139, R140, H141 and F142 within this region are important for maximal binding (12).
Reagent Provided
Anti-TS is provided in liquid form as tissue culture supernatant (containing fetal bovine serum) dialyzed against 0.05 mol/L Tris-HCl, pH 7.2, and 0.015 mol/L sodium azide. Contains stabilizing protein.
