Phosphorylation Site Specific
Reacts with epidermal growth factor receptor (EGFR) phosphorylated at tyrosine residue 1197 (pY1197). Binding of epidermal growth factor to the extracellular domain of EGFR results in receptor dimerization and autophosphorylation on tyrosine residues, pY1197 being one of the major autophosphorylation sites (1, 2).

Monoclonal Mouse Anti-Human EGFR-pY1197, Phosphorylation Site Specific, Clone DAK-H1-1197, is recommended for use in immunohistochemistry. The antibody labels human epidermal growth factor receptor only when, phosphorylated on tyrosine residue 1197 (EGFR-pY1197).

In Western blotting of recombinant phosphorylated human HER1 (EGFR), HER2, HER3 or HER4 protein the antibody labels only the phosphorylated HER1 (EGFR) protein. The antibody does not label non-phosphorylated recombinant human HER1 (EGFR), HER2, HER3 or HER4.
When applied on formalin-fixed, paraffin-embedded cells transfected with either HER1 (EGFR), HER2, HER3 or HER4 phosphorylated intracellular domain constructs the antibody labels only cells expressing the HER1 (EGFR) intracellular domain.
Calf intestinal phosphatase (CIP) treatment of formalin-fixed, paraffin-embedded tissue resulted in loss of staining demonstrating that the antibody recognizes a phosphorylated antigen.
Liquid phase absorption with EGFR-pY1197 peptide prevented staining of formalin-fixed, paraffin embedded tissues. Similar absorption with non-phosphorylated EGFR-Y1197 peptide resulted in strong immunostaining of formalin-fixed, paraffin embedded tissues. This demonstrates that the antibody specifically recognizes the EGFR antigen only when it is phosphorylated on tyrosine 1197.
Database homology searching has shown potential cross-reactivity of the antibody to phosphorylated insulin receptor.
