Polyclonal Rabbit Anti-Human
IgG, Specific for Gamma-Chains
For use in methods demanding a very high specificity. The specificity and performance of the antibody have been ascertained in immunohistochemistry and ELISA. Additionally, the specificity has been tested by crossed immunoelectrophoresis using 12.5 microlitre antibody per square cm gel area against 2 microlitre human plasma.
The F(ab')2 fragment antibody is particularly useful for labeling unfixed blood cells containing active Fc receptors, and for other applications where the Fc part of the antibody molecule could disturb.
Please note that F(ab')2 fragment antibodies are not suited for techniques dependant on aggregation or precipitation of antigen-antibody complexes.

Polyclonal Rabbit Anti-Human IgG, Code A0423, is intended for use in immunohistochemistry. The antibody is also well suited for ELISA and immunoblotting (1).
In immunohistochemistry, the antibody labels plasma cells and related lymphoid cells containing IgG, and it is a useful tool for the classification of patients with B-cell neoplasia (2-4). Additionally, the antibody may be used for distinguishing neoplastic monoclonal proliferation from reactive hyperplasia of plasma cells (2, 5). Differential identification is aided by the results from a panel of antibodies.
Interpretation of results must be made within the context of the patient's clinical history and other diagnostic tests by a certified professional.
The specificity of the antibody has been ascertained as follows:
Crossed immunoelectrophoresis: Only the IgG immunoprecipitation arch appears when using 12.5 µL antibody per cm2 gel area against 2 µL human plasma. Staining: Coomassie Brilliant Blue.
ELISA: No significant reaction is seen in indirect ELISA using human IgA and IgM as coating antigens. In double antibody sandwich ELISA no significant reaction is observed with human plasma stripped of IgG.
Cross-reaction with IgG from other species may occur. As demonstrated by rocket immunoelectrophoresis, the antibody cross-reacts with the IgG-equivalent protein in cat, cow, deer, dog, goat, horse, mink, monkey, mouse, musk ox, polecat, raccoon dog, rat and swine (9).

Dako Human Serum Protein Calibrator, Code X0908, was used as reference for the titre determination. The titre variation between different lots of A0423 is less than 10%. This is achieved by adjusting the titre of each individual lot to match the titre of an antibody reference preparation kept at -80 °C.

For in vitro diagnostic use. F0315 is intended for immunofluorescence, e.g. for the detection of surface IgG on lymphocytes. Being the F(ab')2 fragment of rabbit immunoglobulin, F0315 is particularly well-suited for application on unfixed cells exhibiting Fc receptors, and also for other applications where the Fc part of the antibody molecule could disturb. P0214: HRP-conjugated.
For in vitro diagnostic use. Polyclonal Rabbit Anti-Human IgG/HRP, Code P0214, is intended for the determination of human IgG in ELISA (3, 4) and immunoblotting (5). Additionally, the conjugate is intended for use in immunocytochemistry when IgG is abundant (6). For sensitive detection of IgG in tissues, Dako Polyclonal Rabbit Anti-Human IgG, Code A0423, combined with Dako LSAB+ or EnVision+ visualization systems is recommended. In immunofluorescence, when tested at a dilution of 1:20-1:40 on normal human formalin-fixed, paraffin-embedded tonsil, pretreated with Dako Proteinase K, Code S3020, F0202 gives a strong and specific staining of IgG containing plasma cells.
F0315:
In immunofluorescence, when tested at a dilution of 1:10-1:20 on human formalin-fixed tonsil, a strong and specific staining of IgG containing cells is seen.
P0214:

The Anti-Human IgG conjugate has been produced from a purified immunoglobulin fraction of rabbit antiserum. F0202 is provided in liquid form in phosphate buffer containing 15 mmol/L NaN3, pH 7.2.

Purified immunoglobulin fraction of rabbit antiserum conjugated with horseradish peroxidase (HRP) of veryhigh specific enzymatic activity. The coupling reaction is a modification, developed at Dako, of the two stepglutaraldehyde method of Avrameas and Ternynck. The reaction is gentle, efficient, highly reproducible and gives conjugate molecules of Mr predominantly 200 000 to 240 000. The conjugate is provided in liquid form in 0.05 mol/L Tris/HCI, 15 mmol/L NaN3, pH 7.2.
